The Raji TK+ cell line is a derivative of the Raji cell line, originally established from a Burkitt's lymphoma patient. It is distinguished by its expression of thymidine kinase (TK+), an enzyme critical for nucleotide salvage pathways and DNA synthesis. The Raji TK+ variant is utilized in research areas focusing on cell cycle dynamics, DNA replication, and targeted gene editing techniques. The expression of TK+ makes this cell line a robust model for studying the effects of nucleoside analogs and other TK-dependent therapeutic agents. As a lymphoblastoid cell line, Raji TK+ retains the morphological and immunophenotypic characteristics of its parent line, including high nuclear-cytoplasmic ratio and surface markers typical of B cells, such as CD19 and CD20. It is permissive for Epstein-Barr virus (EBV) studies, which is a hallmark of the Raji lineage, allowing exploration of EBV-related oncogenesis and immune evasion mechanisms. The cell line's genomic stability and reproducibility across experiments further enhance its utility in fundamental and translational research.
